Experimental Studies on the Effects of Tobacco-Smoke-Condensate Application on the Oral Mucous Membrane of the White Rats.


Abstract
To date, many attempts to produce cancer of the oral cavity with various tobacco products have been largely unsuccessful.
In this respect, the author studied experimentally on the effects of tobacco-smoke-condensate application on the oral mucous membrane of the white rats. This expeiment was designed in which extirpation of sublingual gland and needle puncture were achieved prior to applying tobacco-smoke condensate. Experimental animals were killed by cervical dislocation at the intervals of the E, the 10 and the 15 weeks.
Although the fallacy is recognized of attempting to make clinical application of observations made of the experimental aminals, a number of observations may be recorded.
1. In contrast to the group, 5-week A, the group, 5-week B caused comparatively severe tissue changes characterized chiefly by increase in number and chromatin content of basal cells.
2. The group, 10-week B showed severe tissue changes such as focal mitosis and rete peg elongation.
3. The group, 15-week B showed the most severe tissue changes characterized chiefly by prominent nucleoli, increase in N, C ratio, many mitotic figures, the irregular arrangement of basal cells and the presence of epithelial islands, but typical pleomorphism and dyskeratosis were not shown. And also in the group, 15-week A, acanthosis, basal hyperplasia, many mitoic figures and rete peg elongation were manifested.
4. The longer the duration of experiment, the severer was the histopathological change, in all experimental groups.
5. The severity of mucosal change was considerably increased when extirpation of sublingual gland and needle puncture were achieved.
6. In all experimental groups, the gingiva was most severly changed, followed in order by the cheek, tongue, lower lip and palate.
